你干什么？- What are you doing/ What's your job?
你干吗？- What are you doing?! Why are you doing this?!
干嘛？- What's up?!

There are lots of measure words you can use for people. For example.
拨 -bō - 两拨人
伙 - huǒ - 两伙人
队 - duì - 两队人马
条 - 一条女 (广东省这样说)!
群 - qún
名- míng - 两名少年
However these are classifiers used for groups of people, and are not interchangeable. They used with specific nouns. Hope this helps!
